Further studies are clearly required.

Two studies have tested that ability of garlic supplementation to prevent and possibly reverse atherosclerotic plaques. A study on rabbits (Atherosclerosis 1997; 132: 37-42) investigated the effects of Kyolic aged garlic extract on the development of fatty streaks in arteries of rabbits fed high and low cholesterol diets. Fatty streaks are growths of fibrous fatty deposits in the smooth muscle tissue of the arteries and are markers of early atherosclerotic lesions. The study used four groups of animals. Group one received a standard diet, group two a standard diet plus odor free Kyolic, group three a standard diet supplemented with 1% cholesterol, and group four, a standard diet supplemented with 1% cholesterol plus Kyolic.

After six weeks rabbits of group three developed fatty streaks covering 70% of the surface area of the large artery studied (thoracic aorta), which was reduced to 2.5% in the Kyolic treated group four. No fatty streaks were present in groups one and two. The high cholesterol diet in group three caused cholesterol accumulation in the aorta, which was reduced by 50% in the Kyolic supplemented rabbits, in group four. Kyolic also prevented the in vitro growth of smooth muscle cells that occurs in plaque development. The effects were dose related. The higher the dose of Kyolic in the culture medium, the lesser the growth. The study also found that following injury of the carotid artery, Kyolic reduced fatty plaque formation in the intima of the carotid artery by 50%, compared to controls with high cholesterol diet and no Kyolic.

Glutathione is an important defense mechanism in living cells. As a substrate for the antioxidant enzyme glutathione peroxidase, reduced glutathione (GSH) protects cellular constituents from the damaging effects of peroxides formed in metabolism and through other ROS reactions. Decreased tissue GSH levels are associated with cell damage, depressed immunity and the progression of aging, and may increase the risk of cancer development.

The ability of AGE to increase glutathione peroxidase and other ROS scavenging enzymes (Wei and Lau 1998) is important in radioprotection and UV suppression of certain forms of immunity (Reeve et al. 1993), in reducing the risk of radiation and chemically induced cancer (Borek 1993) and in preventing the range of ROS-induced DNA, lipid and protein damage implicated in the disease and aging processes

 

A new Australian study has found that Aged Garlic Extract significantly reduces blood pressure in Australian adults with hypertension, offering hope to those who don’t respond well to prescription medications or would prefer a natural therapy.

 

The study found that two daily capsules of a high potency formula of Aged Garlic Extract was enough to reduce systolic blood pressure by an average of 12 millimetres of mercury – which is comparable to the effect of many commonly prescribed anti-hypertensive medicines.

This is significant because a reduction in systolic blood pressure in this range lowers the risk of cardiovascular disease by 16 – 40 per cent.

Anti-tumor effect. AGE has a variety of pharmacologic effects, including tumor cell growth inhibition and chemopreventative effects. In rodents, AGE and its constituents have been reported to inhibit the development of chemically induced tumors in the bladder (Lau et al. 1986), mammary gland (Amagase and Milner 1993), colon (Sumiyoshi and Wargovich 1990), esophagus (Wargovich et al. 1988), lung (Sparnins et al. 1986), skin (Nishino et al. 1989) and stomach (Wattenberg et al. 1989) in rodents.

 

 

 

 

 

Anti-psychological stress effect. Psychological and physical stresses have been shown to suppress immune functions.  Yokoyama et al. (1986) reported that an AGE preparation containing vitamins B-1 and B-12 restored physical stress–induced immune suppression in mice. Recently, we determined the effect of AGE on immune suppression caused by psychological stress using a communication box. After 4 d of psychological stress, a decrease in spleen weight, spleen cells and hemolytic plaque-forming cells (PFC) was observed in the control mice. AGE significantly prevented the decreases in spleen weight and cells, and restored the reduction of hemolytic PFC caused by the stress
Our results showed that AGE significantly enhanced spermatogenesis after warm water treatment. RGJ, HGJ and DHP, on the other hand, were only slightly effective. Water-administered mice had decreased intromission counts compared with the normal group. AGE significantly improved impotence, whereas RGJ and HGJ were only slightly effective. DGP showed no effect. The effects of AGE were equivalent to those of testosterone. We recently demonstrated that AGE significantly improved peripheral circulation in a cooling rewarming test, which evaluated peripheral circulation. Again, the other three garlic preparations had no effect (Ushijima et al. 1997). The anti-impotence response to AGE may be due to its effects on peripheral circulation. Because oxygen and nutrients are obviously required in spermatogenesis, and it is well known that blood circulation to the testis is poor, improvement in circulation would likely result in improved nutriture and thus spermatogenesis. Furthermore, our previous studies (Ushijima et al. 1997) revealed that AGE significantly improved physical performance, prolonging both active swimming and treadmill running in rodents. Thus, it is possible that AGE might enhance the supply of oxygen to seminiferous tubule cells and stimulate energy metabolism in the testis.

 

AGE inhibits the growth of transplanted tumor cells and enhanced NK and killer cell activities (Kyo et al. 1998).

Recent studies suggest that aged garlic extract (AGE) inhibits formation of AGEPs in vitro and formation of glycation-derived free radicals. S-Allylcysteine, a key component of aged garlic, is a potent antioxidant and can inhibit AGEP formation.

 

AGE protects against atherosclerosis by preventing hypertension, reducing serum cholesterol and triglycerides, and by inhibiting platelet aggregation and LDL oxidation (24,25). AGE also increases activity of antioxidant enzymes such as catalase, superoxidase dismutase, and glutathione peroxidase (26).

 

Recent studies in the author’s laboratory have shown that AGE is an effective inhibitor of AGEPs in vitro (Fig. 3). The key ingredient responsible for the antiglycation properties of AGE was S-allylcysteine, which proved an effective inhibitor of AGEPs (Fig. 4).

swimming time. Subjects given Kyolic were also found to run longer than those given a placebo (control), raw

 

garlic (RGJ), heated garlic (HGJ) or garlic powder (PG) in a mechanical treadmill running test. Control ran for

 

929 seconds whereas those given Kyolic ran for 1611 seconds, almost twice as long.

 

 

Supplementation with the AGE product reduced stress indexes and improved symptoms such as

 

chill, stiffness in the shoulders, fatigue lassitude, decline of physical strength/willpower, headache, abnormal

 

bowel movement, and lumbago. The efficacy of the AGE product was felt by 87.5% of subjects,

 

 

Kawashima et al. (1986) studied 20 healthy male college students and found that Aged Garlic Extract (2 ml,

 

twice per day) improved subjective and objective symptoms of fatigue following 22 days of intense physical

 

training. Aged Garlic Extract also improved patellar reflex and promoted serum levels of GOT, GPT,  triglycerides, and lactic acid, which favor relief from fatigue. These latter effects provided further objective  measurements of the anti-fatigue effects of Aged Garlic Extract. The authors concluded that Aged Garlic Extract  is an effective medicine for the prevention of and recovery from fatigue.
